dc.description.abstract | Candida albicans, es el principal agente causal de la candidiasis vulvovaginal recurrente
(VVCR), las cepas de candida aisladas de pacientes con VVCR presentan una alta resistencia a la
terapia convencionalmente usada para su tratamiento por lo tanto, como alternativa terapéutica
han sido estudiados péptidos antimicrobianos de amplio espectro, dentro de los cuales está la
catelicidina humana LL37.
Este trabajo tuvo como objetivo evidenciar el efecto antifúngico in vitro e in vivo de péptidos
análogos derivados del LL37, en levaduras del género Candida. Para observar este efecto
anti-Candida de los péptidos análogos al LL37, se estudiaron 14 aislamientos clínicos
provenientes de pacientes con VVCR, así como también en 4 cepas de referencia. Se realizó curvas de crecimiento y la técnica de microdilución en caldo, para determinar la concentración
mínima inhibitoria de los péptidos, el fluconazol y los péptidos en sinergia con el fluconazol.
Además se realizó microscopía electrónica de transmisión y de fluorescencia para ver la
afectación estructural de Candida albicans ATCC 10231 frente a los péptidos y por último se
realizó un modelo in vivo de candidiasis sistémica para evaluar la eficacia de los péptidos en
ratones.
Los péptidos análogos al LL37 estudiados presentaron un efecto promisorio anti-Candida y un
destacado efecto sinérgico cuando estos péptidos fueron asociados al fluconazol; a nivel
estructural los péptidos lograron la destrucción de pseudohifas de C. albicans ATCC 10231. Por
lo cual se demostró que estos péptidos se pueden usar como alternativa terapéutica o para
complementar los tratamientos actuales de VVCR. | spa |
